    Method of action of chloroquine

Malaria parasites can enter the body through these mosquito bites, and then live in body tissues such as red blood cells or the liver. This medication is used to kill the malaria parasites living inside red blood cells.

Oct 01, 2018 Mechanism of Action Chloroquine, a 4-aminoquinoline, is an anti-protozoal agent. The precise mechanism by which Chloroquine exhibits activity is not known. Chloroquine, may exert its effect against Plasmodium species by concentrating in the acid vesicles of the parasite and by inhibiting polymerization of heme. It can also inhibit certain enzymes by its interaction with DNA. Another method to prepare chloroquine 3 involves reaction of 83 with methyl acrylate to get via 98 and 99 the adduct 100, which is converted into 7-chloro-1,2,3,4-tetrahydroquinoline-4-one 103. Reaction of 103 with novaldiamine 92 under dehydrogenating conditions gives chloroquine in about 25% overall yield 133 3. Few drugs are available to prevent or treat infections with such parasites and relatively little is known about the modes of action of even such widely used compounds as chloroquine or primaquine. However, much has been learned from investigations of the methods by which the parasites develop resistance to various antimalarials.

Both drugs may be needed for a complete cure and to prevent the return of infection (relapse). In some cases, you may need to take a different medication (such as primaquine) to kill the malaria parasites living in other body tissues.

  3. From a pharmaceutical perspective, I think I would have considered N, SN or Chloroquine. N is hard to get with lots of challenges and opportunities for your efforts not to work. SN is a second option for me due to easy availability but chloroquine appeals to me in it's method of action.

Chloroquine is thought to exert its antimalarial effect by preventing the polymerization of toxic heme released during proteolysis of hemoglobin in the Plasmodium digestive vacuole. The mechanism of this blockade has not been established. We incubated cultured parasites with subinhibitory doses of 3Hchloroquine and 3H quinidine. Two new methods for the simultaneous detn of chloroquine and its two main metabolites monodesethylchloroquine and bisdesethylchloroquine in biol samples, RIA and ELISA, are described. Sensitivity limits are, respectively, 0.70 nM 3 pg of chloroquine sulfate measured in 10 uLof plasma sample for RIA, and 10 nM 22 pg of chloroquine sulfate measured in 5 uL of plasma sample for ELISA. Its mechanism of action is unknown; however, malarial parasites invade human red blood cells, and chloroquine may prevent malarial parasites from breaking down metabolizing hemoglobin in human red blood cells. Chloroquine is effective against the malarial parasites Plasmodium vivax, P. malariae, P. ovale, and susceptible strains of P. falciparum.
